Have it x-rayed.

Weigh it and compare with a full and an empty cartridge.
Laser printers don't have ink, they use toner, which is a powder. As to
"amount", what terms of quantity would be useful? 1/2 full? What does
that mean? Perhaps the unit came with a utility to "estimate" toner
level. On the other hand if you recorded the page count when you
installed it, and note the page count now(printer has to support that
too), and compare the difference to the "rated page count" of the
cartridge, then you may "guess" how full it still is.
