It sounds as if you're trying to launch the command line version of the disk
defragmenter, defrag.exe, from the Start -> Run box. Instead, try entering the
following command.
cmd.exe /k defrag.exe C:
If that's not the case, post back and let us know exactly what you do when
trying to launch the disk defragmenter.
If you want to run the GUI version of the disk defragmenter from the Run box,
enter dfrg.msc. You can also run Disk Defragmenter by right clicking My
Computer, selecting Manage from the menu. There should also be a shortcut on
your Start menu under Accessories -> System Tools.

In Start>Run enter dfrg.msc. Or you can go to Start>Programs>
Accessories>System Tools>Disk Defragmenter. If you use it
often, you can right click Disk Defragmenter and send a shortcut
icon to the desktop.
