Which files should I backup to make a restorable copy of Microsoft Office?
Will it work if I only backup the Microsoft Office program folder, or do I
have to include some other folders?

You can't do this. The only way to make a backup is to make a copy of your
original install media... (CD).
Nothing you suggested will work
You can either image your entire system, or make a copy of the Office cd.
Programs, when installed, scatter files & entries in the windows registry,
in numerous places. The process is 99% of the time impossible to replicate,
other than by installing from origonal media.
